ARP-161 is a human T-cell line, derived from the HUT 78 cell line, infected with non-cloned, non-plaque-purified Simian immunodeficiency virus (SIV). The virus was isolated from a Macaca fascicularis found to be infected at the New England Regional Primate Center.
Note: The virus grows best in CEM×174 cells.
Growth Characteristics: Cells grow as a suspension. Maintain cultures at
0.5 × 106 to 1 × 106 per mL and split twice per week at a ratio of 1:3 to 1:4,
and monitor for virus production. Virus production is dramatic when first
reseeded, then decreases. Aliquots of early passage supernatant should be saved
and stored in liquid nitrogen. When virus production decreases, mix uninfected
CEM×174 cells with infected cells at a ratio of 3:1 or 4:1. Virus production
will increase in 7 to 10 days. Alternately, a thawed 1 mL aliquot of early
passage virus can be used to infect 6 mL of 0.5 × 106 to 1 × 106 CEMx174 cells per mL.
Growth Medium: RPMI 1640 supplemented with
10% fetal bovine serum, 100 U per mL penicillin and 100 µg per mL streptomycin
ARP-161 is negative for mycoplasma,
bacteria and fungi.
Each vial of ARP-161 contains
approximately 9 x 106 cells in RPMI 1640
supplemented 10% fetal bovine serum and 10% dimethyl sulfoxide (DMSO). Please
